What actually happens in the first 72 hours after a treated seed goes in

The commercial argument for seed-applied biology is simple enough that it hides the interesting part. A living organism is placed on a dry seed, stored, transported, planted, and expected to wake up and colonise a root that does not exist yet. Most of the variation in field results comes from that sequence rather than from the strain.

Hour 0 to 6: rehydration

Bacterial cells on the seed coat are in a low-metabolism state. As the seed imbibes, water becomes available at the surface and the cells resume activity. In our qPCR series across 46 sites, viable counts on the seed surface at hour six were on average 82% of the count measured at treatment — but the range ran from 34% to 97%, and the low end correlated almost entirely with two things: contact time with an incompatible fungicide, and storage above 86 °F.

The strain is rarely the variable. Storage temperature and tank partners explain most of the spread we measure at planting.

Hour 6 to 24: the race for the radicle

Once the radicle emerges, exudates begin. This is the signal the inoculant is waiting for, and it is also the signal every other organism in the surrounding soil is waiting for. Colonisation is competitive. A seed placed into a warm, moist, biologically active seedbed gives the applied strains a fair contest; a seed sitting in cold soil below 46 °F for ten days gives them a poor one, because the applied strains are less active at low temperature than several of the native pathogens they are competing against.

Hour 24 to 72: attachment and biofilm

By hour 72, the applied strains have either attached to the root surface and begun forming a biofilm, or they have been outcompeted. Our sampling protocol takes root sections at 72 hours precisely because that is when the outcome is decided. Sites where colonisation density at 72 hours exceeded 10⁵ CFU per gram of root produced a mean yield response of +6.8%; sites below 10⁴ averaged +0.4%, which is inside measurement noise.

What this means for practice

  • Drill treated seed within 14 days. Not because it becomes unsafe, but because counts decline.
  • Check the compatibility table before the treatment, not after. Thiram removes roughly two-thirds of the viable count within four hours.
  • Do not drill into cold soil to hit a calendar date. Below 46 °F the competitive balance moves against you.
  • Store pallets in the shade. A trailer in July sun is the single most common cause of a batch that underperforms without any visible sign.

None of this is exotic. It is the same logic that governs any living input, and it is the reason we publish a count on the delivery note rather than a claim on the label.
